The story takes place in Japan during the Sengoku period, or the Warring States period. Forty-eight major demons, known as majins (lit. demon gods), sense the impending birth of a powerful human, who will grow up to be the vanquisher of demonkind.

故事讲述古日本的室町时代中叶,经历了应任,文明等动乱之后,当权的足利幕府的威信大幅的动摇,是个君不君臣不臣的时代,一名巫师醒糊景光,为了出人头地,而将尚未出生的儿子献祭给48个魔神;一出生就缺少了身上48个部位了幼儿,被狠心的父亲抛入河中,后来被名为寿光的医生所救,替他制作了义眼,义肢,等补足了身上所缺乏的部位,命名为百鬼丸,并抚养他长大.由于百鬼丸的特殊体质,使得他非常容易引来恶灵,将可能危害抚养他的寿光,体验到这点的百鬼丸,于是告别了寿光,出发寻找能让自己安身之处.在寻找魔神的旅途上,百鬼丸遇到了自称大盗的疯疯癫癫的少年.....
